Optimal designs for dose-response models with restricted design spaces

چکیده
In dose response studies, the dose range is often restricted due to concerns over drug toxicity and/or efficacy. We present restricted and unrestricted interval locally optimal designs with respect to a very general class of optimality criteria for estimating the underlying dose response curve. The underlying curve belongs to a diversified set of link functions suitable for the dose response studies and having a common canonical form. These include the fundamental binary response models – the logit and the probit as well as the skewed versions of these models. The results are illustrated through the re-design of a dose ranging trial conducted at the Merck Research Laboratories (Zeng and Zhu, 1997). This work is a generalization of the results of Dai and Zhu (2002) in terms of the design interval, the underlying dose response curve and the optimality criterion. AMS Classification: 62K05, 62J12

Compound Optimal Designs for Percentile Estimation in Dose-Response Models with Restricted Design Intervals
In dose-response studies, the dose range is often restricted due to ethics concerns over drug toxicity and/or efficacy, particularly when human subjects are involved. We present locally optimal designs for the estimation of several percentiles simultaneously on restricted as well as unrestricted design intervals. Our results are applicable to most of the commonly applied link functions with res...
متن کاملBayesian optimal designs for a quantal dose-response study with potentially missing observations.
In a dose-response study, there are frequently multiple goals and not all planned observations are realized at the end of the study. Subjects drop out and the initial design can be quite different from the final design. Consequently, the final design can be inefficient. Single- and multiple-objective Bayesian optimal designs that account for potentially missing observations in quantal response ...

متن کاملSEISMIC OPTIMIZATION OF STEEL MOMENT RESISTING FRAMES CONSIDERING SOIL-STRUCTURE INTERACTION
The main purpose of the present work is to investigate the impact of soil-structure interaction on performance-based design optimization of steel moment resisting frame (MRF) structures. To this end, the seismic performance of optimally designed MRFs with rigid supports is compared with that of the optimal designs with a flexible base in the context of performance-based design. Two efficient me...
متن کاملOptimal designs for comparing curves.
We consider the optimal design problem for a comparison of two regression curves, which is used to establish the similarity between the dose response relationships of two groups. An optimal pair of designs minimizes the width of the confidence band for the difference between the two regression functions. Optimal design theory (equivalence theorems, efficiency bounds) is developed for this non s...
